Description:Sorafenib Tosylate (Bay 43-9006) is a multikinase inhibitor of Raf-1, B-Raf and VEGFR-2 with IC50 of 6 nM, 22 nM and 90 nM, respectively.
Application:Sorafenib act on these cell lines (e.g. Mia PaCa 2, HCT 116, A549, and NCI-H460), inhibition of ERK phosphorylation, causing RAS / RAF abnormal activation pathway. Sorafenib role in MDA-MB-231 breast cancer cell line, completely inhibit the activation of MAPK pathway. Sorafenib inhibit the MEK 1/2 and ERK 1/2 phosphorylation, this effect in a dose-dependent manner, IC50 of 40 nM, respectively, and 100 nM. Sorafenib effect on MDA-MB-231 cells, no effect on the PKB pathway, selective inhibition description Sorafenib MAPK, without inhibiting PKB pathway. Sorafenib effect on the human pancreas (Mia PaCa 2) and colon (HCT 116 and HT-29) tumor cell lines, inhibition of pERK. Sorafenib inhibit the EGFR, IGFR-1, c-met, and HER-2 RTKs, IC50> 10,000 nM. Furthermore, Sorafenibis inhibit VEGFR-2 cell signal. Sorafenib inhibition mVEGFR-2 (flk-1), mVEGFR-3, and mPDGFR-β, IC50 were 15 nM, 20 nM, and 57 nM.
